Reboot. Then try to backup again. This time don't do it from Rom Manager. Try just rebooting to CWM and backup from there. I've had some problems with backing up from Rom Manager.

Make sure you have USB debugging enabled. I think you need that checked, as well.

The only time I've had this problem was do to running out of space on SD card. Make sure you have space for the backup file.

You can't backup with Rom Manager..technically. What you are doing is using the GUI interface of Rom Manager to initiate the Clockworkmod recovery back up/process.
Rom Manager is just a GUI, automated process for downloading ROMs, starting backups, starting ROM installs, etc... It uses Clockworkmod Recovery to do all the processes (install a ROM, make a backup, restore, etc...). It's why Rom Manager says you "need to install clockworkmod recovery" when you try to use it.
clockworkmod recovery is the replacement recovery you can boot to instead of the stock recovery boot image. Rom Manager (if used) will let you click on "Backup my Rom", give it a name and then will reboot the phone into clockworkmod recovery and run the backup routine using the name you gave it in Rom Manager.
